Issue mRNA delivery limits, cold-chain inequity and uncertain regulation for universal vaccine platforms
Summary
mRNA vaccines that require ultra-cold storage are practically impossible to distribute in developing regions without a cold chain, which creates a global health equity problem. Lipid nanoparticle delivery that protects mRNA can trigger allergic reactions to polyethylene glycol and excessive immune responses, leaving limits on safety and stability. Protein vaccines face complex culture and purification processes that restrict production switching and mass manufacture, and mRNA vaccines also struggle with process standardisation and consistent quality at scale. The high mutation rate of RNA viruses shortens the durability of protection and makes regular revaccination necessary. On the policy side, platform technologies lack a unified regulatory definition, approval routes are complex, commercial investment incentives are weak and intellectual property and technology transfer are difficult.
